      Hedgehog(Hh)信号通路及其下游关键基因Patched(Ptch)、Smoothened(Smo)表达异常会引起骨代谢异常。虽然Hh信号通路已被确定为成骨细胞分化和软骨内成骨的重要正调节因子,但其在破骨细胞和骨吸收中的作用机制仍不清楚。最新研究发现,典型Hh信号通路在破骨细胞分化过程中逐渐增强,并且参与破骨细胞的增殖以及凋亡的过程,同时与骨质疏松的发生密切相关。因此,本文拟就Hh信号通路对破骨细胞分化和骨吸收的调控及其分子机制进行综述。

      Abnormal expression of Hedgehog (Hh) signaling pathway and its downstream genes Patched (Ptch), Smoothened (Smo) causes abnormal bone metabolism. Although Hh signaling pathway has been identified as an important positive regulator of osteoblast differentiation and endochondral bone formation, its mechanism of action in osteoclasts and bone resorption remains unclear. Recent studies have revealed that the canonical Hh signaling pathway is gradually enhanced during osteoclast differentiation and is involved in osteoclast proliferation as well as apoptosis, so Hh signaling pathway is closely related to the development of osteoporosis. Therefore, this paper intends to review the regulation of osteoclast differentiation and bone resorption by the Hh signaling pathway and its molecular mechanisms.
